    Diversity Array Technology(DArT)is a hybridization-based genotyping method which uses microarray technology to type and identify DNA polymorphism.DArT has advantages of high quality,high throughput,low cost and the independence on prior sequence information.In this paper,we reviewed the methodology of DArT and improvement on its procedure,application in genetic map construction and crop variety identification.

    Eighty seven accessions of cabbage(Brassica oleracea L.)breeding materials from different geographic origins were evaluated under 26-30 ℃ at the seedling stage for resistance to Fusarium wilt(Fusarium oxysporum f.sp.conglutinans)by using the most virulent isolate from Shanxi Province Shouyang county,named GLHW1,as the inoculum.Type A resistant gene was present in 36 accessions.31 of them were originated from Japan,Korea and some Europe countries,only 2 materials(C59 and C73)belong to traditional Chinese mainland breeding materials.The analysis on distribution of resistant gene in different cabbage populations showed that there were more resistant materials in round cabbage populations introduced from Japan and Korea,and in cabbage materials with grey-green leaves.There was rare resistant gene to Fusarium wilt in Chinese traditional earlymaturity cabbage.The high susceptible materials were amounted to 77.78 %.

    Morphological characterization and evaluation were conducted to assess the genetic diversity in 30 Cucumis sativus L.var.xishuangbannanesis germplasm.Identification of 5 qualitative and 13 quantitative characters suggest that there was relatively abundant genetic diversity in Xishuangbanna cucumber at morphological level.5 quantitative characters related with sex differentiation and lateral branch development showed great difference.Based on above data,the result of cluster analysis expressed that 29 germplasm could be divided into 5 groups,which as basically consistent with important morphological characters.

    Four isozymes:esterase(EST),peroxidase(POD),α-amylase and superoxide dismutase (SOD)extracted from buds of 16 radish(Raphanus sativus L.)accessions were studied with polyacrylamid gel electrophoresis(PAGE).The results showed that a high degree of polymorphism was observed in the EST and POD isozymes.There was a co-dominant locus detected in the zymogram patterns of SOD and α-amylase isozyme,respectively.The isozyme zymogram and cluster analysis revealed genetic diversity and relationship among the tested accessions.This provided valuable theoretical reliance for radish germplasm identification and genetic improvement.

    RsPrx1 cDNA was amplified by reverse transcription-polymerase chain reaction(RT-PCR)from the peel of Chinese red radish(Raphanus sativus L.) root.The full-length of RsPrx1 cDNA was sequenced.Then it was sub-cloned into Pichia pastoris expression plasmid pPIC9KH.RsPrx1 gene was integrated into the genome of Pichia pastoris and successfully expressed by inducing.The recombinants could show certain resistance to NaCl and H2O2.An efficient expression system of Pichia pastoris was constructed,which was the base of large scale production and functional analysis.

    DNA from winter gourd(Benincase hispida Cogn.)was extracted by methods of CTAB,CTAB-Free,PVP,SDS and the effects of different extraction methods were compared and analyzed.By evaluating purity of DNA,concentration of DNA and DNA yield,the result indicated that the effect of CTAB-Free and PVP were better than that of CTAB,SDS.

    Nutritive components of 15 leaves mustard〔Brassica juncea(L.)Czern.et Coss.〕varieties were evaluated and analyzed.The results indicated that there are 3 types leaf mustard varieties,each variety contains 16 kinds of amino acid,among which 7 were essential to human bodies.In those varieties that had higher total amino acid,the contents of essential amino acid,sweet and bitter amino acids were higher relatively.At the same time,the contents of nutritive components of 15 leaves mustard varieties,including VC,calcium,phosphate and iron,etc.There are 1 362-2 428 mg·kg-1(FW)VC,1 002.9-1 872.9 mg·kg-1(FW)calcium,250.7-656.9 mg·kg-1(FW)phosphate and 17.4-32.4 mg·kg-1(FW)iron.

    The processing characteristics of pumpkin(Cucurbita moschata Duch.ex poir.)varieties including carotenoids content,total soluble solid,juicing rate and yield were evaluated comprehensively by the grey correlative degree and clustering analysis.It showed that of all experimental varieties,the correlative degree of‘Beijing No.1’is the closest to that of the given ideal variety,which means‘Beijing No.1’held the better characteristics for juice processing like higher yield,juicing rate,total soluble solid,and carotenoids content.Pumpkin varieties‘Chestnut’and‘Jixiang’can be taken as assistant varieties for improving the quality of pumpkin juice by offering their high total soluble solid and carotenoids content.

    The leaf areas,chlorophyll contents,soluble solid contents and lycopene contents of 17 accessions of pure diploid mini-watermelons(Citrullus lanatus)cultivars and homologous tetraploid watermelons were determined.The results showed that four major agronomic characteristics were increased along with the increase of ploidy.Among which,the leaf areas of tetraploid watermelons were increased by 22.43 %-165.80 %.The chlorophyll a+b contents in the leaf blades of majority homologous tetraploid watermelons were increased 1.57 %-17.28 %,compared with the diploid.And the soluble solid contents in the center of watermelons were increased by 1.80 %-5.69 %.The lycopene contents were increased by 8.66 %-88.41 %,showing significant or extreme significant differences to the diploid.

    Four inbred line of Brassica oleracea var.botrytis,082005,082009,082010 and 082063 were used as paternal plants and male sterile B.rapa ssp.chinensis h08015 was used as maternal plant.Crosses were made to produce inter-specific hybridization through ovary in vitro culture.The results showed that different developmental days of ovary,inoculation method,content of hormone and paternal genotype had significant effects on germination rate of hybrid seed.The ovary in vitro culture could get better results 6 days after pollination.Seed germination rate would be higher than that by other inoculation methods,when seeds were taken out of ovary 4 days after culturing and then continue to be cultured.The MS medium without hormone was better than the MS with 6-BA and NAA,and different paternal genotypes had different appropriate sampling times.To majority paternal genotypes,the best ovary culturing times were 6 days after pollination,but to individual paternal genotype,the appropriate culturing time was 10 days after pollination.

    Taking a wettish-cultivated water dropwort〔Oenanthe stolonifera(Roxb.)Wall.〕new line G06-01 as experimental material,we compared the effects of potassium and planting density on growth of flowering stalk used for propagation.The results showed that the development and germinating rate of axillary bud in unelongated stem of maternal plants were promoted by applying potassium fertilizer,the number of flowering stalk was increased and the axillary bud grew well,the yield was raised in the same planting manner.The average diameter of flowering stalks and their yields were tending to go down at the same time.

    This paper studies the effect on wax gourd(Benincase hispida Cogn.)seed vigor and its physical characteristics treated by salt solution priming with 10 mmol·L-1 CaCl2,NaCl,KNO3,KH2PO4,CaCl2+NaCl(1∶1),KNO3+KH2PO4(1∶1).The results showed that 10 mmol·L-1 KNO3 solution could significantly improve seed vigor and promote seed germination.The seed germination energy,germination percentage,simplified vigor index,activities of SOD and CAT could be raised by 4.0 %,14.7 %,60.0 %,53.2 %,78.7 % respectively,and the electrical conductivity was reduced by 51.2 %。

    Grafting analysis was carried out using three tomato(Lycopersicon esculentum Mill.)lines introduced from Tomato Genetics Resource Center USA(TGRC):LA2701,LA3202 and LA3526 with high resistance to bacterial wilt as rootstocks and‘Zaoguan 30’as cions.Grafted plants were inoculated with Ralstonia solanacearum and bacterial wilt resistance was investigated.Results indicated that the three tomato lines could improve the bacterial wilt resistance obviously.Ralstonia solanacearum in vivo analysis indicated the grafting with bacterial wilt resistant rootstocks could effectively inhibit the propagation of bacterial cells in tomato stem.

    The research used six nitrogen concentrations,they were 0 mg·L-1(T1),94 mg·L-1(T2),140 mg·L-1(T3),210 mg·L-1(T4),315 mg·L-1(T5)and 472.5 mg·L-1(T6).Sweet pepper(Capsicum annuum L.var.grossum Bailey)was taking as experimental material to study the effects of different nitrogen concentration on its seedling ageing formation.The results showed that ageing degree had positive relation with lignin content,whereas the lignin content was affected by nitrogen concentration.Appropriate nitrogen concentration could reduce lignin content,while the higher nitrogen concentration could enhance the lignin content.The lignin content in T6 treatment was 2.09 times of that in T4 treatment.When the nitrogen concentration was 210 mg·L-1(T4),the lignin content in sweet pepper seedling was the lowest.When MDA content reduced,the activities of SOD,POD increased and strong seedling index was higher than other treatments.It indicates when nitrogen concentration is 210 mg·L-1(T4),the anti-ageing strength of sweet pepper seedling is increased.

    Preliminary sampling investigation on wild vegetable resources in Nyingtri,Tibet was carried out.The results indicated that wild vegetables in Nyingtri were abundant.There were 71 species,belonging to 31 families and 53 genus.They could be divided into 44 kinds of leaf vegetables,10 of stem vegetables,8 of root vegetables,4 of fruit vegetables,2 of flavouring and 3 of other types according to their edible organs.

    Zigan No.2’is a new red cabbage F1 hybrid developed by crossing a CMS line CMSSD13 as female parent with an inbred line ZGX02 as male parent.It’s a mid-early maturing variety,taking about 70 days from transplanting to harvest.The round head is compact and tough in structure which makes it a long shelf life.Its leaf head is 15 cm in height,15 cm in diameter,with a short core of 6.5 cm in length.The head weight is 1.5-2.0 kg.The anthocyanin content is 547.6 mg·kg-1(FW).It has higer resistance to TuMV and black rot than‘ziyun’.Its yield is about 75 t·hm-2 and it is suitable for cultivation in spring or autumn.

    ‘Zhenong No.6’is a new vegetable soybean variety developed by crossing‘Taiwan 75’as female parent with‘9806’as male parent.It is limited setpod,36.5 cm in plant height.Its node numbers of main stem is 8.5,and the effective branch number is 3.7.Its growing period is about 86 days,and it is 4 days lower than‘Taiwan 75’.Its pods are green with few and white down,the length and width of pod with 2.0 grain are 6.2 cm and 1.4 cm,respectively.The weight of a hundred fresh pod and a hundred fresh grain are 294.2 g and 76.8 g.The starch content is 5.2 %,the total soluble sugar content is 3.8 %.The fresh pod yield is about 10.5 t·hm2.

    Zaojiang No.2’is a new early maturing variety of cowpea developed by crossing early maturing variety‘Zaojiang No.1’as female parent with‘Zhijiang 14’as male parent.The first harvest can reach 24.0 t·hm-2,the total yield is about 49.5 t·hm-2.At present,the total extended areas in Henan,Shandong,Shanxi,Jiangsu,Gansu,Liaoning are 8 333 hm2.

    Yumo No.1’is a new konjac variety which was systematically selected from a series of plants came from Yunnan province.It grows about 130 days.The triennium plant is 84.5 cm in height,68.9 cm in width.The leafstalk is 45.7 cm in length and 2.5 cm in diameter.The green laminae are pinnate split or second pinnate split,the stalk is cyan and with black spot,the corm is round with snuff color and white flesh.It has excellent characteristics such as dry matter content is about 21.5 %-22.2 %,konjac glucomannan content is about 59.8 %-60.2 %.Its yield is 30 t·hm-2.It is suitable to be cultivated in southwest of China.
