Abstract:Using CTAB method, template DNAs were extracted from young roots of germinated wax gourd(Benincasa hispida Cogn.)seeds. Electrophoresis revealed that DNA bands extracted were clear and bright, indicating that the DNA quality could suit RAPD analysis. PCR amplification profiles of black rind wax Gourd variety-‘Heilaocu’ F1 and its parents were analyzed by using 140 RAPD primers. Only one primer, SPS-C15 was found to generate stable and complementary special DNA fragment. A production seed sample was identified with this primer, showing 90.7 % of seed purity. While the seed purity was 89.8 % in field test. The results of these two methods were consistent, suggesting that RAPD technology could be used for rapid testing of seed purity in wax gourd F1 hybrid.
