Cloning and Sequence Analysis of Tomato spotted wilt virus Coat Protein Gene in Yangling Region of Shaanxi Province

Tomato spotted wilt virus (TSWV) is one of the top 10 harmful viruses in the world.In order
to determine the situation of tomato infected by TSWV in Yangling region of Shaanxi,this experiment collected
4 suspected TSWV infected tomato samples from this region,and carried out phenotypic observation and PCR
molecular identification.The results showed that 2 samples had amplified a specific band of 234 bp,indicating
that they were infected by TSWV.The coat protein gene(CP)of 757 bp TSWV in Yangling region was obtained
by homologous cloning method,which had the highest homology with Spanish TSWV isolate,reaching 99%,but
had a distant relationship with Kunming TSWV isolate.
