Abstract:According to literature report and the design of primers registered by GenBank,a quantitative
RT-PCR(RT-qPCR)detection technology system for potato virus Y(PVY)was established.There existed
a favorable linear correlation(R2 = 0.991 2)between this technical standard curve of cycle threshold and
template concentration.This system could be used to detect PVY specifically,and the sensitivity was 1 000
times of the conventional RT-PCR and yet with good repeatability.PVY in potato plants from Shandong,
Qinghai,Guizhou,Heilongjiang,and Hebei 5 provinces,were detected by the established detection method.
The detection rates were 16.00,33.33,25.00,14.29 and 20.00 percentage point higher than that of the
conventional RT-PCR technology,respectively.Therefore,the established PVY RT-qPCR detection system
had the characteristics of strong specificity,high sensitivity,quick and accurate detection of PVY contents
from diseased potato seeds and plants.This study had provided effective technical means for early monitoring
and effective control of potato virus Y.
