番茄立枯病菌拮抗菌B4-8的鉴定及生防作用评价

Identification of Strain B4-8 Against Rhizoctonia solani and Evaluation of Its Biocontrol Effect

摘要: 为了明确对番茄立枯病有生防效果的菌株B4-8 的分类地位,并探索其生防作用机理和安全性,基于形态特征、生理生化特性及16S rDNA 序列分析,对菌株B4-8 进行鉴定;以平板对峙培养法测定其抑菌谱,并通过盆栽试验评价其对番茄立枯病的生防效果;采用生理生化和分子生物学的方法初步分析菌株的作用机制,并以洋葱和生菜叶中脉致病模型及PCR扩增检测毒力基因BCESM(Burkholderia cepacia epidemic strain marker)的方法测定该菌株对植物和人的安全性。结果显示,拮抗菌株B4-8 为洋葱伯克氏菌(Burkholderia cepacia)。该菌株对番茄立枯病菌、苦瓜枯萎病菌、柑橘沙皮病菌等6 种病原菌都具有较强的抑制活性,对番茄立枯病的室内防治效果达71.43%。该菌株可抑制番茄立枯病菌菌丝的生长,生物膜形成能力较强,可产生蛋白酶、几丁质酶以及硝吡咯菌素等抑菌代谢物质。菌株B4-8 中未检测到毒力基因BCESM,不能使洋葱和生菜叶中脉致病,初步表明其对植物和人体安全,具有防治番茄立枯病的潜力。

Abstract: In order to clarify the taxonomic status of strain B4-8 having biocontrol effect on tomato damping-off,and explore its biocontrol mechanism and safety for human being,this study carried out identification on strain B4-8 based on morphological,physiological and biochemical characteristics and 16S rDNA sequence analysis.The antifungal spectrum was determined by plate confrontation culture method,and its biocontrol effect on tomato damping-off was evaluated by pot experiment.The strain action mechanism was preliminarily analyzed by physiological,biochemical and molecular biological methods.The safety of strain B4-8 for plants and human being was determined by onion and lettuce leaf midrib infection models,and PCR amplification detecting gene BCESM(Burkholderia cepacia epidemic strain marker ).The results showed that the antagonistic strain B4-8 was Burkholderia cepacia.This strain had stronger inhibitory activity on 6 pathogenic fungi,including Rhizoctonia solaniFusarium oxysporum and Deightoniella torulosa,etc..Its indoor control efficacy on tomato damping-off reached 71.43%.This strain could inhibit mycelial growth of tomato damping-off;had stronger ability of forming biofilm;could produce antimicrobial metabolites,including protease,chitinase and nitropyrrolidin,etc..However,BCESM gene was not detected in strain B4-8 by PCR,and strain B4-8 was unable to cause pathogenicity of onion and lettuce leaf midrib,preliminarily indicating that it was safe for plants and human being,and had potential to control tomato damping-off.
