Using Seed Protein SDS-PAGE to Identify Heat-resistant Radish Germplasm

利用种子蛋白SDS-PAGE技术进行抗热萝卜种质鉴定

Abstract: Seed proteins of 32 heat-resistant radish(Raphanus sativus L.)accessions were analyzed with sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)to estimate their polymorphism.A total of 41 protein bands were detected,of which 25(61 %)were polymorphic.Each accession could produce 28 to 39 protein bands,their profiles were different with each other and all accessions could be distinguished.These accessions could be clustered into two major groups of white and red roots at similarity index of 0.76 with UPGMA.The protein band similarity coefficient ranges from 0.64 to 0.98 and the average is 0.79.The results revealed that cluster analysis of seed protein was highly in accordance with radish root color,and low genetic diversity and narrow genetic basis existed in these heat-resistant radish accessions.

摘要: 运用SDS聚丙烯酰胺电泳(SDS-PAGE)技术,对32份抗热萝卜材料种子蛋白的遗传多态性进行研究,共检测出41条蛋白谱带,多态性谱带25条(61 %);每份材料可检测出28~39条谱带,所有材料蛋白谱带均有差异,可进行品种鉴别;供试材料蛋白谱带相似系数为0.64~0.98,平均为0.79;聚类分析表明,在相似系数0.76处所有种质材料可分为白皮与红皮两大类。研究结果显示,基于种子蛋白谱带的聚类结果与肉质根皮色有较高的相关性,抗热萝卜材料遗传多样性较低,遗传基础比较狭窄。
