Screening of RAPD Tagging and SCAR Conversion Related to Aphid Resistant Gene in Pepper |

Abstract In this study, F2 plants from the cross combination of aphid resistant pepper(Capsicum annuum L.)cultivar 03-C79′, and aphid infection A0003 were used to screen the molecular marker linked to aphid resistant gene in pepper with RAPD molecular marker and BSA(bulked segregation analysis)method. RA750, a specific primer of polymorphism, was obtained from screening 200 RAPD primers for a pepper with aphid resistant traits. The marker from the results of PCR was tested in individual DNA reaction for F2 population and the RA750 amplified fragment could only be detected in the aphid resistant plants. The distance between RA750 and gene was 6.03 cM. According to the sequence of fragment analysis, one pair of primers had been designed. The RAPD marker was converted into a SCAR marker which was named RA750-S.
