DING Zijian,ZHANG Junhua,WU Lili,TANG Xiaoling,JIANG Qingqing,CHENG Weitao,YANG Bozhi,ZHOU Shudong
In order to further improve the anther culture efficiency of pepper, taking pod pepper XR1 as the experimental material, the eff ects of pretreatment of fl ower buds at normal temperature(25 ℃,CK)and low-temperature(0、4、8 ℃)on the efficiency of XR1 anther culture and changes in physiological indicators were studied. The results showed that compared with normal temperature, after low-temperature pretreatment of flower buds, the anther browning rate was significantly reduced, and the induction rate of embryoids was significantly increased. In addition to the significantly reduced MDA content in anthers, the contents of stress-resistant substances(soluble proteins, soluble sugars,Pro)and the activities of antioxidant enzymes (POD,PPO)were signifi cantly increased. The anther browning rate was the lowest under 0 ℃ treatment, the induction rate of cotyledon-shaped embryoids was the best under 4 ℃ treatment, and the induction rate of non-cotyledon-shaped embryoids was the best under 8 ℃ treatment.Among the anthers, except for the highest MDA content under 0 ℃ treatment, the other physiological indicators reached the highest values under 4 ℃ treatment.Correlation analysis showed that there was a negative correlation between the anther browning rate and the induction rate of embryoids(cotyledon-shaped and non-cotyledon-shaped),and there was an extremely signifi cant negative correlation with the MDA content and the strongest correlation, with a correlation coeffi cient of -0.967.The induction rates of both cotyledon-shaped and non-cotyledon-shaped embryoids were extremely significantly positively correlated with the soluble sugar content in the resistance index and had the strongest correlation, with correlation coefficients of 0.837 and 0.784,respectively.Only cotyledon-shapedembryoids could induce tissue culture seedlings during anther culture, and the pretreatment of flower buds at 4 ℃ had the best eff ect.The research results can provide a theoretical basis for in-depth analysis of the benefits of low-temperature pretreatment in improving the efficiency of pepper anther culture.

SHE Xiaoling,WANG Shaoli,ZHOU Chengxian,WU Dan,HE Jiting,LYU Yang,LIU Lijuan
Melon aphid(Aphis gossypii)is an important limiting factor for facility watermelon production.During field survey,we discovered in spring facility cultivation there was a certain amount larva of Allothrombium ovatum .Whether these larvae could inhibit the population of melon aphid? It was currently unclear. This study conducted monitoring on larva of Allothrombium ovatum and contemporaneous population of melon aphid ,in 2 watermelon plastic-covered tunnels without chemical application at Yanqing district of Beijing;and also analyzed their occurrence dynamics. The results showed that in Yanqing plastic-covered tunnel and watermelon fields the main occurrence stage for Allothrombium ovatum was from May 19th to June 15th.These mites were either free crawling or parasitic on the body of melon aphids. In 2 watermelon plasticcovered tunnels the naturally occurring mite strain rates could reach as high as 26.67% and 33.33%.The number of mites per hundred stains could reach 83.33 heads and 100 heads.Meanwhile, the highest rate of aphid was 26.67% in the same period. Subsequently, the melon aphid populations declined. The exponential population growth trend under suitable environment was not yet presented, indicating that in spring facility watermelon plastic-covered tunnel Allothrombium ovatum could suppress the population of melon aphid in suitable climate years.Attention should be paid to the protection and utilization in production.
